Treatment of heart failure remains a daunting task due to the limited regenerative capacity of the heart. Accordingly, cardiac repair using stem cells or engineered cardiac grafts has gained increased attention. Current stem cell treatment only leads to a mild increase in cardiac function as the majority of the injected stem cells don't survive and only few stem cells differentiate into cardiomyocytes. In embryo's, cardiac cell growth and differentiation are tightly controlled by growth factors and their specific cardiac niche, including electrical and mechanical load, to evolve in functional cardiac tissue. In this project we will create a model system, the bio-artificial cardiac niche, in which human mesenchymal stem cells can be subjected to coordinated biological and physical (mechanical and electrical) stimuli within a 3D environment. This approach will enable to study physiological mechanisms required for optimal cell preconditioning and creation of tissue engineered "pro-cardiac" constructs.
