Project summary: Currently, there is a growing demand for effective enzymes that can be implemented in existing or new industrial processes, e.g. for replacing chemical reagents. Although the use of enzymes as biocatalysts is much "greener" than conventional chemical procedures, natural enzymes are typically only able to act on substrates present in nature. Therefore, creating enzymes that are able to convert novel substrates by enzyme engineering methodology represents a big challenge and is of considerable biotechnological importance. In the current STW-project we aim at creating industrially relevant oxidative enzyme by a novel directed evolution approach. Briefly, this approach involves the construction of huge randomized enzyme libraries followed by the selection of desired enzymatic activities using any target substrate. The main challenge is to develop ultra-high-throughput screening techniques that enable to screen massive enzyme collections.
