Omschrijving van het onderzoek

Autoimmune diseases such as rheumatoid arthritis (RA), multiple sclerosis (MS) and type 1 diabetes are characterized by the presence of autoantibodies in the blood of the patient. An early diagnosis of the disease, which is so important for earlier and more effective treatment, depends very often on the sensitive detection of such disease-specific autoantibodies. The autoantibodies are often directed towards normal cellular proteins containing a modification that is introduced after translation. Serological detection of autoantibodies in type 1 diabetes is currently performed using either purified native proteins or recombinant variants of possible antigens. These methods are suboptimal since the accessibility and/or the purity of the proteins is often a problem. More important, however, is the fact that often particular posttranslational modifications are needed for the autoantigenicity of these proteins, which is very difficult to achieve at the protein level. The aim of this project is to identify posttranslationally modified molecular structures that are specifically recognized by type 1 diabetes autoantibodies, which can be applied in the development of peptide-based diagnostic tests.
