The discovery of antibiotics has revolutionized medicine, extending human life span. However, the rapid development of antimicrobial resistance and the concurrent decline in the discovery of new drugs calls for innovative drug discovery efforts. Soil microorganisms are a rich source of untapped antimicrobials that has remained largely unexplored due to a lack of technologies to comprehensively mine strain collections. Lipopeptides constitute a promising class of antibiotics, with daptomycin as the last new drug class to enter the clinic. In this project, we will discover and derivatize new lipopeptide antibiotics related to daptomycin and polymyxins using state-of-the-art computational genomics, chemical elicitation, metabolomics, and synthetic chemistry, for pre-clinical development. Thus, we will provide solutions for the key societal and clinical challenges posed by the emergence of polymyxin resistance and the nonapplicability of daptomycin in lungs, to the potential benefit of patients around the globe.
